Description
The Kdm6a gene encodes a histone demethylase that catalyzes the demethylation of tri/dimethylated lysine 27 of histone H3. These mice possess loxP sites on either side of exon 24 of the targeted gene. Homozygous females and hemizygous males (this gene is X linked) are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exon 24 deleted in the cre-expressing tissues.

When female mutant mice are bred to a strain with widespread Cre recombinase expression (see Stock No. 003724), or to a strain with Cre recombinase in spermatids (see Stock No. 003328), this mutant mouse strain may be useful in studies of development.

Development
A FRT site flanked targeting vector containing a NEO selection cassette and a 3' loxP site was utilized in the construction of this mutant. This selection cassette was inserted downstream of exon 24 of the targeted gene, and another loxP site was inserted upstream of exon 24. This construct was electroporated into (C57BL/6N x 129S6/SvEvTac)F1 derived iTL1 BA1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts. The resulting chimeric animals were crossed to knock in mice (on the 129S4 genetic background) expressing Flp recombinase driven by the Gt(ROSA)26Sor promoter (Stock No. 003946). Mice that retained the loxP site flanked exon 24 were then bred to remove the Flpe recombinase allele. Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.

"MICE" means mouse strains, their progeny derived by inbreeding or crossbreeding, unmodified derivatives from mouse strains or their progeny supplied by The Jackson Laboratory ("JACKSON"). "PRODUCTS" means biological materials supplied by JACKSON, and their derivatives.
